Next-gen intranasal Measles/Mumps-vectored vaccine platform for robust protection against SARS-CoV-2 and RSV

PROBLEM

SARS-CoV-2, particularly the emerging Omicron variants, and respiratory syncytial virus (RSV) propose significant threat to global public health. Both SARS-CoV-2 and RSV are highly transmissible and significant risk of disease in infants and young children, older adults, and immunocompromised individuals. Current vaccines, administered intramuscularly, generate robust systemic neutralizing antibodies, but mucosal immunity in the respiratory tract is insufficient, thus failing to control viral transmission. As a result, current vaccines reduce severe disease but offer limited prevention of infection and transmission, leaving high-risk populations vulnerable, thereby highlighting the critical need for vaccines that will bolster both mucosal and systemic immunity to prevent spread of SARS-CoV-2 and RSV.

SOLUTION

Researchers at The Ohio State University and Nationwide Children’s Hospital have developed novel intranasal multivalent SARS-CoV-2 and RSV vaccines using attenuated measles virus (MeV) and mumps virus (MuV). Recombinant MeV and MuV vectors were generated to express SARS-CoV-2 Omicron spike protein, RSV F proteins from both A and B strains, and full-length RSV G protein, thus generating constructs formulated into multivalent vaccines that target both viruses simultaneously.
