Novel Antibodies Targeting IL-2 for Treatment of CancerProblem IL-2 and its receptor IL-2R are known to play multiple roles in shaping the immune response, including both immune activation and tolerance. Treatments activating effector cells against tumors are undermined by an autoinhibitory loop caused by endogenous IL-2 secreted from activated Teffs. Their dual roles in shaping the immune response present a complex landscape for clinical exploitation. Current IL-2 therapeutics cannot address the negative feedback loop, resulting in endogenous IL-2 stimulating Treg expansion, and limiting efficacy. There is a need for a new treatment to provide adequate clinical response and immune modulation for conditions associated with IL-2 signaling, such as cancer and autoimmune disorders. Solution Dr. Mark Rubinstein and team have developed novel IL-2 antibodies to bind human IL-2, modulating its signaling pathways and improving clinical outcomes. These antibodies have differential effects on IL-2 binding to its receptors, offering targeted therapy options. With high affinity and specificity, these novel IL-2 antibodies inhibit IL-2 signaling through various receptor chains. The antibody inhibits IL-2 signaling through IL-2 Rαβγ to a greater extent than through IL-2 Rβγ. When complexed with IL-2 prior to infusion into mammalian host, IL-2/anti-IL-2 mAb complexes have much stronger biological activity and half-life than IL-2 alone. Applications
